[장선희 기자 = 경기북부/강원] 을지대학교의료원 혈액내과 김동욱 교수와 울산과학기술원 김홍태 교수, 경희대학교 이윤성 교수팀이 만성골수성백혈병의 급성기 진행에 결정적 역할을 하는 SH3BP1, PACSIN2, COBLL1 등 새로운 유전자 네트워크를 발견했다고 밝혔다.

 

만성골수성백혈병은 적혈구·백혈구·혈소판 등 혈액세포를 만드는 조혈줄기세포가 BCR-ABL1유전자 발생에 의해 비정상적인 혈액세포를 폭발적으로 늘리는 과정에서 발생하는 질환이다. 

 

백혈병 환자는 지난 2001년 세계 최초의 표적항암제 이매티닙(글리벡)이 사용된 이후 다양한 표적항암제 개발로 장기 생존과 완치에 대한 희망을 얻게 됐지만, 일부 환자에서 약물 내성이 발생하거나 급성기로 진행될 경우 1년이내 사망하게 된다. 

 

김 교수 연구팀은 COBLL1 단백질을 중심으로 만성골수성백혈병 급성기 진행 및 약물 내성 관련 유전자 네트워크를 분석한 결과, 주변 단백질인 SH3BP1, PACSIN2의 발현 증감이 백혈병의 진행과 표적항암제에 대한 내성을 조절하는데 긴밀하게 연관되어 있음을 밝혔다.

 

김동욱 교수는 “이번 연구를 통해 백혈병 전체의 약제 내성과 급성기 진행을 조기에 진단할 수 있는 진단키트와 표적항암제 개발이 한층 더 빨라질 것으로 기대한다”고 말했다.

 

김 교수와 연구단이 발표한 이번 연구결과는 Cancer Medicine 학술지 3월호에 게재됐다. 

 

COBLL1 단백질은 만성골수성백혈병 환자에게 표적항암제 약물 내성을 유발해 급성으로 진행되게 영향을 미치는 중요한 유전자로, 2017년 전세계 최초로 김 교수 연구팀이 규명해 백혈병 분야의 세계적인 권위지 ‘루케미아(Leukemia)’에 실렸다.

 

이를 계기로 을지대학교(의정부을지대학교병원)와 울산과학기술원(UNIST), 연세대학교, 고려대학교, 경희대학교 등 13개 기관 및 바이오·제약기업은 지난 5년간 백혈병 진단 및 치료 신기술 개발을 위해 의생명과학, 나노공학, 인공지능 기술을 융합한 세계적 수준의 산-학-연-병 백혈병 연구 네트워크를 구축해 올 2월 ‘백혈병 초정밀바이오 연구단’을 출범했다. 연구단은 이번 연구와 같이 지속적인 공동 연구 및 성과발표를 통해 주목받을 것으로 기대된다.

 

또 을지대학교의료원 백혈병오믹스연구소는 단독으로 연구한 만성골수성백혈병 표적함암제 치료 3개월차 골수를 이용한 염색체 검사와 말초혈액 정량유전자증폭검사(RQ-PCR)를 상호보완적으로 활용하여, 국제치료표준지침인 ELN 재정 및 만성골수성백혈병의 치료 지침을 제시하는 논문을 발표했다.

 

해당 연구 결과는 만성골수성백혈병 치료 초기에 정확한 판단을 통해 장기간의 치료 방향을 제시할 수 있을 것으로 평가되어 지난 2021년 11월 Leukemia Research에 게재됐다.
